3T-MRI analysis of epidermis and dermis moisturizing using the T2-mapping sequence

Abstract: T2-mapping in MRI has been applied to assess basal free water content of the epidermis and dermis and to search for an increase in free water content after applying a moisturizing cream. The T2-value and the proton density (PD)-weighted signal of the epidermis, superficial, and deep dermis of the calf were measured using a T2-Mapping sequence first without cream and then at intervals after applying moisturizing cream on three 4 x 4 cm square regions of interest from top to bottom at the posterior aspect of the calf. After application of moisturizing cream, there was no significant difference in T2-value of the epidermis and superficial dermis at 1, 3, and 6 h. T2-value increase in the deep dermis was statistically significant at 1 h (+27.5%, P = 0.037), and at 6 h (+21.3%, P = 0.051), but not significant at 3 h. The PD-weighted signal increased significantly in the epidermis at 6 h (+22.4%, P = 0.048), superficial dermis at 6 h (+31.8%, P = 0.028), and deep dermis at 1 h (+22.4%, P = 0.042). Skin 3T-MRI using T2-mapping sequence can yield a cartography of the water distribution of skin layers, with the PD-weighted more sensitive than the T2 value.
