The most important aspect of any HPLC method development scheme is to achieve adequate resolution among all analytes. This requires evaluation and optimization of several chemical and physical parameters. This presentation will demonstrate how pH, mobile phase composition, column bonded phase and temperature can be manipulated to provide the best overall separation for both isocratic and gradient modes of LC separation. There will be particular emphasis on utilizing mobile phase pH to optimize peak shape and resolution for ionizable components. Attendees will leave this meeting with an understanding of the chemistry that affects the mobile phase-sample-bonded phase interaction. This knowledge will allow chromatographers to develop better methods in less time whether they follow a systematic manual process or a computer-aided approach to method development. This will reduce the time needed to produce rugged, robust HPLC methods.

Method development has traditionally been a long, tedious activity and chromatographers continually attempt to reduce the time and effort required to develop an optimal LC method. Efficient method development can be realized by two factors - understanding the effect of method parameters on chromatography and automating the necessary evaluations in creating "the best" LC method. In the past decade, HPLC column technologies have undergone significant development driven by needs for high throughput, high sensitivity and column stability/lifetime requirements. With new and improved LC phases and formats continually being introduced, there are an overwhelming variety of columns from which to choose for optimized method development. In this presentation, we will discuss recent column technologies, their benefit in method development and guidance in selecting the appropriate column. Of particular focus will be the recent advances in high resolution, high speed chromatography by implementing various columns for LC analysis, such as the sub-two micron- and superficially porous particles, monolith technologies and phase chemistries such as Eclipse Plus.
